本发明涉及肺癌靶向CHRNA5基因的shRNAs表达载体的构建、筛选及其用途。本发明利用RNA干涉技术，针对CHRNA5基因的不同靶序列，以pLLU2G为载体，构建了两个能在哺乳动物细胞中表达shRNA的表达质粒CHRNA5-shRNA1/2。本发明肺癌靶向CHRNA5基因的shRNAs表达载体能高效的特异性的抑制CHRNA5基因表达，用于制备治疗高表达CHRNA5基因的肺癌的基因药物。
