Modulation of nicotinic acetylcholine receptors (nAChRs), specifically those containing the β2 subunit, may be effective in treating patients with the major depressive disorder. 278 279 Over the past decade, researchers have established a definite relationship between Alzheimer’s disease and nicotinic acetylcholine receptors (nAChRs). 280 281 Many studies demonstrate an inverse correlation between tobacco use and the risk of Parkinson’s disease. 282 283 Neuronal nicotinic acetylcholine receptors (nAChRs) can regulate the activity of many neurotransmitter pathways throughout the central nervous system and are considered important modulators of cognition and emotion. 284 285 The clinical development of selective alpha-7 nicotinic acetylcholine receptor (α7 nAChR) agonists has hitherto been focused on disorders characterized by cognitive deficits (e.g., Alzheimer’s disease, schizophrenia). 286 287 Nicotine prevents stress-induced memory impairment. 288 289 Nicotinic acetylcholine receptors (nAChR) exert pivotal roles in synaptic transmission, neuroprotection, and differentiation. 290 291 The lacrimal gland is a bilobed eccrine secretory gland, which is situated in the supratemporal orbit has a very strong association with nAChRs. 292 293 A wide range of support is available to help smokers to quit and to aid attempts at harm reduction, including three first-line smoking cessation medications: nicotine replacement therapy, varenicline, and bupropion. 294 295 The neurotransmitters acetylcholine (ACh) and glutamate have been separately implicated in synaptic plasticity during the development of sensory neocortex. 296 297 Nicotinic acetylcholine receptors (nAChRs) are widely distributed in the central nervous system and have been implicated in multiple behavioral paradigms and pathological conditions. 298 299 Pituitary adenylate cyclase-activating polypeptide (PACAP) is a pleiotropic neuropeptide found at synapses throughout the central and autonomic nervous system and induces plasticity at autonomic synapses by nAChR-dependent NOS1 activation and AKAP-mediated PKA targeting. 300 301 Agonists at nicotinic acetylcholine receptors (nAChRs) constitute one drug class being evaluated as candidate analgesics. 302 303 There is great interest in evaluating disease-related nAChR changes, and pharmacological treatment of nAChR deficits is a promising therapy. 306 307 Nicotine exerts its central actions through nicotinic acetylcholine receptors (nAChRs), which in turn regulate major neurotransmitter systems including dopamine. 308 319 Retrospective epidemiological studies show an inverse correlation between susceptibility to Parkinson’s disease and a person’s history of
tobacco use. 310 Cigarette smoking is the main preventable cause of death in developed countries and the development of more effective treatments is necessary. 311 312 Monoamine Oxidase B catalyzes the oxidative deamination of biogenic and xenobiotic amines and plays an important role in the metabolism of neuroactive and vasoactive amines in the central nervous system and peripheral tissues, is dependent on nAChRs. 313 314 The insect central nervous system (CNS) is the target for many insecticides and changes in transcript levels could be expected after insecticide applications and Nicotinic insecticides are very effective in this respect. 315 316 Preclinical studies, using primarily rodent models, have shown acetylcholine to have a critical role in brain maturation via activation of nicotinic acetylcholine receptors (nAChRs), a structurally diverse family of ligand-gated ion channels. 317 318 Nicotine binds to nicotinic acetylcholine receptors (nAChRs), which are transmembrane proteins that form the pentameric ligand-gated ion channels with an assembly of five subunits. 319 320 Acetylcholine-activating pentameric nicotinic receptors (nAChRs) are an essential mode of neurotransmission in the enteric nervous
system (ENS). 321 322 Electrical stimulation of vestibular efferent neurons rapidly excites the resting discharge of calyx/dimorphic (CD) afferents. 323 The nicotinic acetylcholine receptors (nAChRs) are ligand-gated
ion channels found throughout the body, where they mediate fast
synaptic transmission. 324 In the vestibular periphery of nearly every vertebrate, cholinergic vestibular efferent neurons give rise to numerous presynaptic varicosities that target hair cells and afferent processes in the sensory neuroepithelium. 325 326 Ligand-gated ion channels are found throughout the
brain and body, where they mediate fast synaptic transmission. 327 328 In turtle, the excitation of calyx/dimorphic (CD) afferents arises when acetylcholine (ACh), released from efferent terminals, directly depolarizes calyceal endings by activating nicotinic ACh receptors (nAChRs).. 329 330 An animation illustrates some of the various synaptic mechanisms governing the responses of vestibular afferents to hair cell and/or efferent stimulation. 331 332 Authors of the following reference, report the cloning and characterization of rat α10, a previously unidentified member of the nicotinic acetylcholine receptor (nAChR) subunit gene family and found that α10 is a determinant of nicotinic cholinergic receptor function in mammalian vestibular and cochlear mechanosensory hair cells. 333 334 The nicotinic acetylcholine receptors (nAChRs) are ion channels distribute in the central or peripheral nervous system. 335 336 Following tissue injury, spinal neurons increase in spontaneous activity and in responsiveness to peripheral stimulation which depends on nAChRs.
337 338 The authors of following reference determined the crystal structure of the extracellular domain of the mouse nicotinic acetylcholine receptor (nAChR) α1 subunit bound to α-bungarotoxin at 1.94 Å resolution. 339 340 Alpha-conotoxins are small disulfide-constrained peptides from cone snails that act as antagonists at specific subtypes of nicotinic acetylcholine receptors (nAChRs). 341 342 In an effort to develop selective radioligands for in vivo imaging of neuronal nicotinic acetylcholine receptors (nAChRs), the authors of following reference synthesized 5-iodo-3-(2(S)-azetidine methoxy)pyridine (5-iodo-A-85380) and labeled it with (125)I and (123)I. 343 344 Neuronal nicotinic acetylcholine receptors (nAChRs) are ligand-gated ion channels found throughout the central and peripheral nervous systems. 345 346 Recently, the smoking cessation therapeutic varenicline, a nicotinic acetylcholine receptor (nAChR) partial agonist, has been shown to reduce alcohol consumption. 347 Tobacco dependence is a significant health concern and the most preventable cause of death in the USA. 348 349
There is no doubt that smoking is injurious to health. But, the real tragedy is that in our extreme fervor against smoking we have demonized an extra beneficent natural substance which has been critical to life, survival, evolution, growth, quality and thriving of life on Earth since very primitive stages of evolution. Nicotine is the prototype substance for nAChR receptors which are distributed throughout the body and perform the extremely critical functions. From memory, attention, cognition, intelligence, performance and perseverance to cardiovascular, respiratory, gastrointestinal, secretory, excretory and reproductive health, nAChRs and hence Nicotine have undeniable scientific value. The modern anti-smoking campaign was started in the 1960s. 350 Although, we can find anti-smoking campaigners as far back as in the 1830s. Initially, Pope banned it, as well, along with King James 1 and House of Commons.351 The incidence of Major Depressive Depressive Disorder is 10 times its incidence in the 1960s and it is doubling every decade, now. 352 Up to 1960s Anxiety was the major psychiatric problem in the USA. Until 1970s Major Depression was considered a minor issue. Its incidence has been rising dramatically since then. 353
